Pemirolast is a small molecule mast cell stabilizer used as an anti-allergic drug, primarily for the prevention and treatment of ocular symptoms such as itching associated with allergic conjunctivitis. It works by inhibiting the release of inflammatory mediators (including histamine and leukotrienes) from human mast cells, thereby preventing Type I immediate hypersensitivity reactions. Pemirolast also inhibits chemotaxis of eosinophils into ocular tissue and blocks mediator release from eosinophils. Its mechanism includes antagonism at the histamine H1 receptor and inhibition of antigen-stimulated calcium influx into mast cells. Pemirolast has been studied for other indications such as asthma and was considered in computational studies for potential disruption of SARS-CoV-2 spike protein interaction with ACE2 receptors[1][2][5][7].
