Overview

Penicacid W is a novel mycophenolic acid derivative isolated from the marine-derived fungus *Penicillium senticosum* RCDB005, which was discovered in sediment samples from the South China Sea. It belongs to a series of newly identified compounds (penicacids O-W) characterized for their anti-proliferative properties. In preclinical evaluations, Penicacid W (identified as compound 9 in research) demonstrated significant inhibitory activity against human cancer cell lines, particularly the MOLM-13 acute myeloid leukemia (AML) cell line, with IC50 values in the micromolar range (0.13 to 1.13 μM). As a derivative of mycophenolic acid, its mechanism of action likely involves the inhibition of inosine-5'-monophosphate dehydrogenase (IMPDH), an enzyme essential for de novo guanosine nucleotide biosynthesis, thereby suppressing the proliferation of malignant cells.
