Overview

Penpulimab + anlotinib is a combination therapy consisting of two distinct agents used primarily in oncology. Penpulimab is a novel anti-PD-1 monoclonal antibody engineered with IgG1 subclass heavy chains and Fc segment modifications, which enhance stability, reduce aggregation, and lower the risk of infusion-related adverse effects compared to traditional IgG4 PD-1 antibodies. It blocks the programmed cell death protein 1 (PD-1) pathway, thereby enhancing T-cell mediated immune responses against tumors[3]. Anlotinib is a small molecule multi-target receptor tyrosine kinase inhibitor that suppresses angiogenesis and tumor proliferation by inhibiting VEGFR, FGFR, PDGFR, c-Kit, and downstream PI3K/AKT signaling pathways[1][3]. The combination has demonstrated promising efficacy and manageable safety in several cancers including small cell lung cancer (SCLC), unresectable hepatocellular carcinoma (uHCC), recurrent/metastatic head and neck squamous cell carcinoma (R/M HNSCC), advanced hepatocellular carcinoma (HCC), and rare subtypes like pulmonary giant cell carcinoma[2][3][4][5][6][7].
