Overview

ACU-D1 is a topical small molecule drug that acts as a first-in-class inhibitor of the 26S proteasome. It is being developed primarily for the treatment of moderate to severe rosacea. The active ingredient is pentaerythritol tetrakis (3-(3,5-di-tert-butyl-4-hydroxyphenyl) propionate). In clinical trials, ACU-D1 demonstrated significant efficacy in reducing inflammatory lesions and erythema associated with rosacea. Its mechanism involves inhibition of the 26S proteasome and NF-kappa B signaling pathways, which are implicated in inflammation. The drug has shown a favorable safety profile in phase II studies and may also be under investigation for HPV-associated lesions in HIV-positive individuals[1][2][4][5][7].
