PENTOCLO

PENTOCLO is a combination drug regimen consisting of pentoxifylline (a methylxanthine derivative), tocopherol (vitamin E), and clodronate (a bisphosphonate). It is primarily used for the medical management of osteoradionecrosis (ORN) of the mandible and other bones following radiotherapy for head and neck cancers. Pentoxifylline improves blood flow by decreasing blood viscosity and has anti-inflammatory effects; tocopherol acts as an antioxidant to reduce oxidative stress; clodronate inhibits bone resorption by osteoclasts. The combination aims to reduce radiation-induced fibrosis, promote mucosal and bone healing, and decrease the need for surgical intervention in refractory ORN cases[1][2][5][6]. Clinical studies have shown that long-term use can lead to significant symptom improvement, reduction in exposed bone area, mucosal healing, pain relief, and overall better outcomes compared to conventional therapies[1][2].
