Overview

Pentolinium is a ganglionic blocking agent used primarily as an antihypertensive drug. It acts as a nicotinic acetylcholine receptor antagonist, specifically targeting neuronal nicotinic receptors (notably alpha 3 beta 4 subtypes) at autonomic ganglia. By binding to these receptors, pentolinium inhibits the release of noradrenaline and adrenaline from adrenergic nerves, leading to smooth muscle relaxation and vasodilation. This mechanism results in lowered blood pressure and makes it useful for managing acute or chronic hypertension, particularly during surgery or hypertensive crises. Pentolinium is typically administered orally, intramuscularly, or intravenously[1][3][4].
