Overview

Pentoxifylline + tocopherol is a combination therapy investigated for its potential anti-fibrotic effects in patients with chronic hepatitis C. Pentoxifylline is a non-selective phosphodiesterase inhibitor that improves blood rheology and possesses anti-inflammatory properties, specifically by inhibiting the production of tumor necrosis factor-alpha (TNF-alpha) and potentially modulating transforming growth factor-beta (TGF-beta) signaling. Tocopherol (Vitamin E) is a lipid-soluble antioxidant that helps mitigate oxidative stress, a key driver of hepatic stellate cell activation and subsequent collagen deposition. The French National Agency for Research on AIDS and Viral Hepatitis (ANRS) sponsored a Phase 3, double-blind, placebo-controlled trial (NCT00119119) to evaluate whether a 12-month regimen of this combination could reduce liver fibrosis in patients who were non-responders or intolerant to standard interferon-alfa and ribavirin therapy.
