Pentoxifylline + vitamin E is a combination therapy consisting of pentoxifylline, a non-selective phosphodiesterase inhibitor, and vitamin E (tocopherol), an antioxidant. Pentoxifylline increases intracellular cyclic AMP (cAMP) and cyclic GMP (cGMP), improves red blood cell and leukocyte membrane flexibility, and enhances microvascular blood flow. Vitamin E acts as an antioxidant, reducing oxidative stress in tissues. The combination has been studied primarily for the prevention and treatment of radiation-induced fibrosis and other late side effects following radiotherapy or chemoradiotherapy in cancer patients. Preclinical studies have shown that this combination can reduce tissue fibrosis by more than 50%, likely through downregulation of TGF-β and TNF-α pathways[1][2][3][5]. Clinical trials have demonstrated its efficacy in reducing radiation-induced fibrosis, lymphedema, and soft tissue injury[3][4][5].
