Pentrys is an anti-idiotypic p53 peptide-based immunotherapy (vaccine) that was developed for the treatment of hormone-refractory prostate cancer (HRPC). The drug consists of specific peptides derived from the p53 tumor suppressor protein, which is frequently overexpressed or mutated in advanced prostate cancer. Pentrys is designed to be administered intradermally in combination with granulocyte-macrophage colony-stimulating factor (GM-CSF), which acts as an adjuvant to stimulate T-cell responses against p53-expressing tumor cells. In Phase II clinical trials, the vaccine was found to be well-tolerated and capable of inducing anti-p53 seroconversion in a subset of patients; however, it demonstrated minimal clinical activity and failed to produce significant PSA responses, leading to the cessation of its development.
