Overview

PEP-CMV is an "off-the-shelf" peptide-based cancer vaccine designed to target the human cytomegalovirus (CMV) antigen pp65, which is frequently expressed in various malignant tumors such as glioblastoma and medulloblastoma, but absent in healthy brain tissue. Developed at Duke University and licensed to Annias Immunotherapeutics, the vaccine consists of a 26-amino acid synthetic long peptide (SLP) that encodes multiple Class I, Class II, and antibody epitopes. It is administered as an emulsion in the adjuvant Montanide ISA 51. The treatment protocol typically involves pre-conditioning with a tetanus/diphtheria toxoid and lymphodepletion with temozolomide to enhance the immune response. PEP-CMV is being investigated for the treatment of recurrent malignant glioma, medulloblastoma, and newly diagnosed glioblastoma in both pediatric and adult populations.
