Pep19 is a synthetic peptide, sequence DIIADDEPLT, derived from a peptide naturally present in human cells. It acts as an **inverse agonist of the cannabinoid type 1 receptor (CB1R)**, part of the endocannabinoid system, which is critical in regulating metabolism, appetite, fat breakdown, and energy balance. Clinical studies show that oral Pep19 reduces visceral fat (up to 17% in obese adults without affecting lean mass) and significantly improves sleep quality, with no significant adverse effects. Its suggested mechanism of anti-obesity action in both animal and human studies involves **browning of white adipose tissue** and increasing UCP1 gene expression, facilitating increased energy expenditure. Preclinical studies report additional metabolic benefits such as improved insulin sensitivity, reduction in hepatic fat, improved blood sugar, cholesterol, and blood pressure. Pep19 is orally administered, is not allergenic or mutagenic, carries GRAS (Generally Recognized as Safe) status, and is being studied as a potential alternative for patients intolerant to GLP-1-based and conventional weight loss therapies.
