Drug intelligence / Profile preview
Pep2-mut (also known as Pep-Nur77R346K) is an 18-amino acid cell-permeable peptide derived from the orphan nuclear receptor Nur77 (NR4A1) containing an R346K mutation. Developed by researchers at the Affiliated Drum Tower Hospital of Nanjing University Medical School, Pep2-mut acts as a molecular decoy that targets the PRMT5/Nur77 methylation axis. It selectively disrupts the interaction between Nur77 and AKT, thereby suppressing the inhibitory phosphorylation of Nur77 at serine 351 (S351) while preserving its endogenous symmetric dimethylarginine (SDMA) methylation at arginine 346 (R346) mediated by protein arginine methyltransferase 5 (PRMT5). This restores the proper chromatin occupancy of Nur77 and promotes endometrial stromal cell (EnSC) decidualization. Pep2-mut is being investigated for the treatment of female infertility, specifically recurrent implantation failure (RIF) and embryo implantation failure.
