Overview

**PeptiCRAd-1** is a clinical-stage, peptide-coated conditionally replicating adenoviral cancer immunotherapy developed by Valo Therapeutics. It comprises the oncolytic adenovirus VALO-D102, engineered to express the immune costimulatory ligands CD40L and OX40L, with its capsid coated with tumor-antigen peptides derived from NY-ESO-1 and MAGE-A3. Following intratumoral administration, the virus is intended to selectively replicate in tumors, induce local inflammation and tumor-cell lysis, and prime systemic antigen-specific cytotoxic T-cell responses against tumors expressing NY-ESO-1 and/or MAGE-A3. It is being evaluated in the Phase 1 START study, including in combination with pembrolizumab, for injectable advanced solid tumors.
