Peptide-33 is a senomorphic peptide discovered through a deep learning-driven platform designed to identify novel sequences with anti-aging potential. In preclinical studies using human skin fibroblasts, Peptide-33 demonstrated protective effects against UVB-induced damage, reduced reactive oxygen species (ROS) levels, and decreased senescence-associated beta-galactosidase (SA-β-gal) staining. Its mechanism of action involves a dual pathway: the upregulation of the Vascular Endothelial Growth Factor A (VEGFA)–VEGFR2 signaling pathway to enhance tissue regeneration and the suppression of the mechanistic Target of Rapamycin (mTOR) signaling to modulate stress responses and cellular senescence. It is being investigated as a next-generation intervention for skin aging and inflammatory aging in dermatology.
