Overview

Peptide A49 is a synthetic therapeutic peptide developed by researchers associated with Assistance Publique - Hôpitaux de Paris (AP-HP) for the treatment of glioblastoma. It is derived from the C-terminal domain of thrombospondin-1 (TSP-1) and functions as a CD47 agonist. Unlike CD47-blocking antibodies that aim to promote macrophage-mediated phagocytosis by disrupting the CD47-SIRPα "don't eat me" signal, Peptide A49 directly triggers a caspase-independent form of programmed cell death (PCD). This mechanism involves the induction of mitochondrial depolarization, the exhaustion of ATP, and the accumulation of reactive oxygen species (ROS), effectively bypassing the apoptotic resistance commonly found in aggressive brain tumors like glioblastoma. Preclinical studies have demonstrated its efficacy in inducing cell death in both glioblastoma cell lines and patient-derived stem cells.
