Drug intelligence / Profile preview
Peptide-I is a cell-penetrating peptide developed by researchers at Peking University First Hospital to treat myocardial infarction (MI) complicated by stress hyperglycemia (SHG). Under hyperglycemic conditions, the interaction between the transcription factor FOXK2 and O-GlcNAc transferase (OGT) is promoted, leading to site-specific O-GlcNAcylation of FOXK2 at threonine 552, which triggers its polyubiquitination and proteasomal degradation. Peptide-I acts by disrupting the FOXK2–OGT interaction, thereby stabilizing FOXK2, restraining macrophage inflammatory activation (specifically suppressing the IL-6/STAT3 signaling axis), and improving post-infarction cardiac repair and function.
