The peptide-pulsed cDC1 vaccine is an investigational autologous cell-based cancer immunotherapy consisting of conventional type 1 dendritic cells (cDC1s) loaded (pulsed) ex vivo with MHC class II tumor-associated peptides derived from oncogenic drivers (such as HER2 and HER3). Developed primarily by the H. Lee Moffitt Cancer Center and Research Institute, the vaccine is designed to be administered intrathecally directly into the cerebrospinal fluid to activate CD4+ Th1 adaptive immune responses against cancer cells. It is being evaluated in a Phase 1 clinical trial (NCT05809752) for the treatment of breast cancer-associated leptomeningeal disease (LMD), including HER2-positive and triple-negative breast cancer (TNBC) subtypes.
