Peptide vaccine + Poly-ICLC is a combination immunotherapy consisting of a synthetic or tumor antigen-derived peptide-based cancer vaccine administered together with the immunostimulatory adjuvant Poly-ICLC (polyinosinic-polycytidylic acid stabilized with polylysine and carboxymethylcellulose). The peptide vaccines are designed to elicit tumor-specific T cell responses by presenting selected tumor-associated or neoantigen peptides, often restricted by HLA type, to the immune system. Poly-ICLC acts as a Toll-like receptor 3 (TLR3) agonist, enhancing innate and adaptive immune activation, promoting dendritic cell maturation, and boosting antigen-specific cytotoxic T lymphocyte responses. This combination has been evaluated in multiple clinical trials for various cancers including glioblastoma multiforme (GBM), diffuse midline gliomas (including DIPG), breast cancer, melanoma, pancreatic cancer, and others. The approach aims to improve anti-tumor immunity by combining targeted vaccination with potent immune stimulation[1][4][5][6][8].
