Overview

Peptide YY is a 36-amino acid gut hormone, also known as PYY or peptide tyrosine tyrosine, secreted by L-cells in the ileum and colon after eating. It exists mainly in two forms: PYY(1–36) and the truncated form PYY(3–36), with the latter being the predominant circulating form. Peptide YY acts primarily to reduce appetite and food intake by binding to neuropeptide Y receptors, especially neuropeptide Y receptor type 2 (Y2R), in both peripheral tissues and the central nervous system. Its mechanism involves modulation of hypothalamic circuits that regulate satiety. Peripheral administration of PYY has been shown to decrease caloric intake in humans and animals, making it a target for obesity therapy research[1][2][4][5]. Additional physiological effects include inhibition of exocrine pancreatic secretion, vasoconstriction, and reduced gastrointestinal motility[7]. Clinical development has focused on its potential for treating obesity; however, side effects such as nausea have limited its therapeutic use so far[6].
