PER2 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the Period circadian protein 2 (PER2) gene. PER2 is a core component of the mammalian circadian clock and plays a significant role in regulating inflammatory responses. Research indicates that PER2 can promote inflammation by upregulating pro-inflammatory cytokines such as TNF-α and IL-6 and suppressing the anti-inflammatory activity of BMAL1. In preclinical studies, PER2 siRNA is used to knockdown PER2 expression in cell lines (such as RAW 264.7 macrophages) and animal models to investigate its role in chronic inflammatory skin disorders like atopic dermatitis. Silencing PER2 has been shown to reduce the production of inflammatory cytokines and nitric oxide, suggesting that targeting the circadian clock components may offer a therapeutic pathway for managing allergic and inflammatory diseases.
