Perenostobart is a humanized IgG4 monoclonal antibody that selectively targets the ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1, also known as CD39) ectoenzyme on tumor and immune cells, thereby inhibiting the conversion of extracellular ATP/ADP to AMP and reducing downstream immunosuppressive adenosine production in the tumor microenvironment.[1][4][5][6] By blocking CD39 enzymatic activity, it is intended to enhance antitumor immune responses and is being developed for the treatment of advanced solid tumors, including pancreatic, gastric, lung, and other malignancies, primarily in early-phase clinical trials as monotherapy and in combination with chemotherapy and checkpoint inhibitors.[1][2]
