Overview

Perifosine + paclitaxel is an investigational drug combination consisting of perifosine, an oral alkylphospholipid and inhibitor of the AKT pathway, and paclitaxel, a widely used antineoplastic agent that stabilizes microtubules and disrupts mitosis. Preclinical studies show that the combination synergistically induces apoptosis in various cancer cell lines, including ovarian and non-small cell lung cancers, by inhibiting AKT/mTOR signaling, activating pro-apoptotic pathways (such as caspase 3 and JNK), increasing ceramide and reactive oxygen species (ROS) production, and suppressing cellular glycolysis and mitochondrial metabolism. This combination has been studied mainly for its ability to overcome resistance mechanisms in cancer cells, particularly those refractory to standard chemotherapies.[1][4]
