perifosine + sorafenib

**Perifosine + sorafenib** is an oral combination regimen of two small molecule inhibitors: perifosine, an AKT inhibitor, and sorafenib, a multikinase inhibitor that targets RAF/MEK/ERK pathway (as well as VEGFR, PDGFR, and other kinases). This drug combination has demonstrated synergistic antitumor effects in preclinical models, particularly by concurrent inhibition of the PI3K/AKT and RAF/MEK/ERK pathways, resulting in enhanced blockade of cell proliferation, mitochondrial dysfunction, induction of necroptotic and apoptotic cell death, and tumor regression. Preclinical studies support investigation of this combination for the treatment of relapsed/refractory Hodgkin lymphoma and hepatocellular carcinoma, with early phase clinical trials also conducted for advanced solid tumors including renal cell carcinoma[1][2][3][5][6][4].
