**Perifosine + tamoxifen** is an investigational combination therapy of two small-molecule drugs—perifosine, an oral alkylphospholipid that inhibits Akt signaling pathway, and tamoxifen, a selective estrogen receptor modulator (SERM) used primarily for estrogen receptor-positive (ER+) breast cancer. This combination is being evaluated for its potential to overcome resistance to endocrine therapy, particularly in ER+ metastatic breast cancer. Tamoxifen acts by antagonizing estrogen activity at the estrogen receptor, thus inhibiting proliferative signaling in breast tissue. Perifosine inhibits Akt phosphorylation and disrupts downstream survival and proliferative pathways, which are frequently implicated in cancer cell resistance to hormone therapies. The combination is tested for synergistic or additive anti-tumor effects and possibly more durable suppression of tumor growth compared to monotherapy. Perifosine has been developed primarily by AEterna Zentaris, while tamoxifen is a well-established generic pharmaceutical compound.
