Overview

Peripheral blood stem cell transplantation (PBSCT) in this context refers to the intervention arm of the BMT CTN 0201 Phase III randomized trial, led by the Medical College of Wisconsin. The procedure involves the administration of granulocyte colony-stimulating factor (G-CSF), such as filgrastim or lenograstim, to healthy HLA-compatible unrelated donors to mobilize hematopoietic stem cells from the bone marrow into the peripheral circulation. These cells are then collected via apheresis and infused intravenously into patients with hematologic malignancies, including acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), chronic myeloid leukemia (CML), myelodysplastic syndromes (MDS), and myeloproliferative disorders. The goal of the transplant is to achieve full hematopoietic and immune reconstitution in the recipient following myeloablative conditioning. The BMT CTN 0201 trial specifically compared the outcomes of PBSCT against traditional bone marrow transplantation to evaluate survival and graft-versus-host disease (GVHD) rates.
