Overview

Peripheral blood stem cells (PBSC) are hematopoietic progenitor cells collected from the donor's blood after mobilization (typically with G-CSF). In the context of the HAMLET trial (NCT04366206) led by DKMS gemeinnützige GmbH, these cells are used for allogeneic hematopoietic cell transplantation in patients with high-risk acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), or myelodysplastic syndromes (MDS). The study compares outcomes between transplants from partially matched unrelated donors (MMUD) and haploidentical donors, both utilizing post-transplant cyclophosphamide (PTCy) to mitigate graft-versus-host disease (GvHD). The primary therapeutic goal is the restoration of the recipient's hematopoietic system and the induction of a graft-versus-leukemia (GvL) effect to prevent disease relapse.
