Periplaneta americana aqueous extract fraction 3-10 kDa (PAW2) is a biologic fraction derived from the American cockroach (*Periplaneta americana* L.) that is being investigated as a potential therapeutic agent for ulcerative colitis. This specific fraction is obtained through sequential ultrafiltration of aqueous extracts from *Periplaneta americana*, specifically retaining molecules with a molecular weight between 3 and 10 kDa, which include bioactive peptides, proteins, and polysaccharides. In preclinical studies using DSS-induced murine models of ulcerative colitis, PAW2 demonstrated significant anti-inflammatory effects by inhibiting the TLR4/MyD88/NF-κB signaling pathway and reducing the production of pro-inflammatory cytokines such as TNF-α, IL-17A, and IFN-γ. Additionally, PAW2 helps preserve intestinal barrier integrity by upregulating tight junction proteins, including occludin-1 and ZO-1, and modulates the composition of the gut microbiota to alleviate dysbiosis.
