Periplocin is a natural cardiac glycoside monomer isolated from the root bark of *Periploca sepium* Bunge (Cortex Periplocae), a traditional Chinese medicine. It is being investigated for its potent anti-tumor properties, particularly in the treatment of ovarian cancer. Periplocin's mechanism of action involves the restoration of 5-hydroxytryptamine receptor 1E (HTR1E) expression and its associated anti-tumor signaling. It achieves this by competitively binding to the CCCTC-binding factor (CTCF), which releases CTCF from the topologically associating domain (TAD) boundary between HTR1E and ZNF292. This epigenetic modulation inhibits the proliferation and peritoneal dissemination of ovarian cancer cells. Additionally, periplocin has been studied for its cardiotonic effects and its ability to induce apoptosis in various other cancer types.
