Overview

Perkileucel is an autologous tumor-infiltrating lymphocyte (TIL) adoptive cell transfer (ACT) therapy being developed by the East Metropolitan Health Service in Australia. It is designed for the treatment of patients with unresectable stage III or metastatic melanoma who have progressed on prior anti-PD-1-containing immune checkpoint inhibitor therapy. The therapy is manufactured by harvesting a patient's own TILs from surgically excised melanoma lesions, expanding them ex vivo, and then reinfusing them as a single intravenous dose. The treatment protocol is preceded by a 5-day lymphodepleting chemotherapy regimen consisting of cyclophosphamide and fludarabine, and is followed by up to six doses of high-dose aldesleukin (interleukin-2) to support T-cell survival and expansion in vivo.
