Overview

Peroxiredoxin 2 (PRDX2) is a thiol-specific antioxidant enzyme that reduces peroxides to alcohols, playing a key role in cellular redox homeostasis and antioxidant defense. In the context of Crohn's disease (CD), PRDX2 has been identified as an anti-fibrogenic factor that is downregulated in patients with intestinal strictures. Research indicates that PRDX2 expression can be induced by the metabolite inosine via the adenosine 2A receptor (A2AR). PRDX2 acts by reducing cellular stress and inducing cyclooxygenase-2 (COX-2), which subsequently inhibits collagen expression in human ileal fibroblasts. Therapeutic approaches under investigation include lentiviral-mediated gene overexpression of PRDX2 to ameliorate intestinal fibrosis, as demonstrated in preclinical SAMP1/YitFc mouse models.
