Drug intelligence / Profile preview
pERP1-ADC (also known as MZB1-ADC) is a preclinical-stage antibody-drug conjugate (ADC) targeting plasma cell-induced ER resident protein 1 (pERP1, also known as MZB1). Developed by researchers at the Cleveland Clinic, the ADC consists of a monoclonal antibody directed against the human pERP1 antigen conjugated to an unspecified cytotoxic payload. pERP1 is a co-chaperone protein specifically required for the differentiation and function of antibody-secreting cells, and is highly expressed on the surface of multiple myeloma cell lines and patient-derived CD138+ plasma cells. In preclinical studies, pERP1-ADC demonstrated potent in vitro cytotoxicity against the human myeloma cell line RPMI18226 and significantly prolonged survival in a transgenic multiple myeloma mouse model (AEY-PVM) at a dose of 1.25 mg/kg.
