Personalized Combination Drug Therapy (Swedish Medical Center) is an individualized therapeutic strategy developed by the Ivy Brain Tumor Center at Swedish Medical Center for patients with recurrent glioblastoma multiforme (GBM). This approach involves isolating cancer stem cells (CSCs) from a patient's own tumor and performing high-throughput screening (HTS) against a library of approximately 73 FDA-approved compounds. These compounds span various therapeutic classes, including antineoplastics, anti-infectives, and NMDA receptor antagonists. Based on the screening results, a personalized cocktail of up to three drugs demonstrating the highest potency and potential for synergy against the patient's specific tumor cells is selected and administered. This Phase 0/1 study aims to evaluate the safety and preliminary efficacy of tailoring medication cocktails to the unique biological characteristics of an individual's recurrent GBM, rather than using a standardized treatment regimen.
