Moonshot Antibodies is developing a personalized monoclonal antibody cocktail therapy that targets patient-specific tumor neoantigens identified through genomic sequencing. This investigational treatment uses directed evolution in yeast and phage display systems to engineer multiple human monoclonal antibodies specific for an individual patient's tumor neoepitopes. These antibodies are combined into a customized cocktail designed to overcome tumor heterogeneity and resistance by simultaneously targeting multiple tumor-specific mutations. The mechanism of action involves recruiting the immune system to destroy cancer cells through antibody-dependent cellular cytotoxicity (ADCC) and other effector functions. The therapy is currently in Phase 1 development for malignant melanoma, triple-negative breast cancer, and colorectal cancer, and is often intended for use in combination with checkpoint inhibitors.
