A personalized neoantigen mRNA vaccine is an individualized cancer immunotherapy designed to stimulate a patient's immune system to recognize and attack tumor cells based on their unique mutational profile. The production process involves sequencing a patient's tumor and healthy tissue to identify tumor-specific mutations, known as neoantigens, which are not present in normal cells. mRNA sequences encoding these specific neoantigens are then synthesized and typically encapsulated in lipid nanoparticles (LNPs) for delivery. Once administered, the mRNA is translated by host antigen-presenting cells into neoantigenic peptides, which are presented on MHC molecules to activate a targeted T-cell response. In the context of the ChiCTR2500095931 trial, this vaccine is being evaluated by the Third People's Hospital of Yancheng as an adjuvant therapy in combination with the PD-1 inhibitor camrelizumab and platinum-based chemotherapy for patients with resectable non-small cell lung cancer (NSCLC) following curative-intent surgery.
