Overview

A personalized neoantigen vaccine platform developed by Washington University School of Medicine for the treatment of pediatric patients with recurrent or refractory brain tumors. The vaccine is tailored to each patient by identifying unique tumor mutations (neoantigens) through exome and cDNA-capture sequencing of tumor and normal tissue. It is designed to target up to 20 prioritized patient-specific neoantigens to stimulate a robust T-cell mediated immune response. The platform encompasses both polyepitope DNA plasmid vaccines, typically delivered via electroporation (e.g., using the TDS-IM v2.0 device), and synthetic long peptide vaccines. These vaccines are generally administered in combination with the adjuvant poly-ICLC (Hiltonol), a Toll-like receptor 3 (TLR3) agonist, to enhance dendritic cell activation and overall immunogenicity.
