Overview

Pertuzumab is a recombinant humanized monoclonal antibody targeting the extracellular dimerization domain (subdomain II) of the human epidermal growth factor receptor 2 (HER2). It acts as a HER dimerization inhibitor by binding specifically to HER2 and blocking its heterodimerization with other HER family receptors, thereby inhibiting tumor cell growth. Pertuzumab also mediates antibody-dependent cellular cytotoxicity. It was developed by Genentech and is now marketed by Roche after their acquisition of Genentech. Approved in 2012 by the US FDA for treatment of HER2-positive metastatic breast cancer, it is used in combination with trastuzumab and chemotherapy agents such as docetaxel. Clinical trials have demonstrated significant improvement in progression-free survival and overall survival in patients with HER2-positive breast cancer.
