HLX-11 is a biosimilar candidate of pertuzumab (Perjeta), a recombinant humanized monoclonal antibody developed by Shanghai Henlius Biotech and licensed to Organon for global commercialization. It targets the extracellular dimerization domain (Subdomain II) of the human epidermal growth factor receptor 2 (HER2). By binding to this domain, HLX-11 inhibits the heterodimerization of HER2 with other HER family receptors, such as HER1 (EGFR), HER3, and HER4, thereby blocking downstream signaling pathways like MAPK and PI3K that promote cell proliferation and survival. It is primarily being developed for the treatment of HER2-positive breast cancer in neoadjuvant, adjuvant, and metastatic settings.
