Overview

This combination therapy is a neoadjuvant regimen being evaluated in a Phase II clinical trial sponsored by Yale University for the treatment of clinical stage I-II HER2-positive breast cancer. The regimen consists of two HER2-targeted monoclonal antibodies, pertuzumab and trastuzumab, administered alongside a chemotherapy backbone known as T-FEC. T-FEC includes weekly paclitaxel followed by a combination of 5-fluorouracil, epirubicin, and cyclophosphamide. Pertuzumab works by binding to the extracellular dimerization domain II of the HER2 protein, preventing it from pairing with other HER family members, while trastuzumab binds to domain IV. The addition of these targeted agents to standard chemotherapy aims to improve pathologic complete response (pCR) rates in patients with early-stage HER2-positive breast cancer by providing comprehensive blockade of HER2 signaling while utilizing cytotoxic agents to induce cell death.
