pertuzumab-DLP1 is an experimental antibody-drug conjugate (ADC) designed to target the human epidermal growth factor receptor 2 (HER2). It consists of the monoclonal antibody pertuzumab conjugated to a proprietary dual-linker payload known as DLP1. The DLP1 payload is distinctive for its dual mechanism of action, combining a topoisomerase I inhibitor and a microtubule inhibitor within a single conjugate. This approach is intended to maximize cytotoxic efficacy and overcome resistance mechanisms, such as payload insensitivity or heterogeneity, that often limit the effectiveness of single-payload ADCs like trastuzumab deruxtecan (Enhertu). In preclinical studies, pertuzumab-DLP1 has demonstrated potent anti-tumor activity across various HER2-expressing cancer models, including breast, gastric, and colorectal cancers, and has served as a critical benchmark for the development of next-generation biparatopic ADCs like TJ106.
