Pertuzumab zuvotolimod (SBT6050) is a first-in-class immune-stimulating antibody conjugate targeting HER2, consisting of the humanized anti‑HER2 monoclonal antibody pertuzumab covalently linked via a linker to a potent small‑molecule toll‑like receptor 8 (TLR8) agonist. By binding HER2 on tumor cells, the conjugate localizes the TLR8 agonist to the tumor microenvironment, where it activates myeloid cells and downstream NK and T cells, while the pertuzumab component antagonizes HER2 signaling and mediates Fc‑dependent effector functions. Initially developed by Silverback Therapeutics for HER2‑expressing solid tumors, it entered phase 1/2 trials in advanced HER2‑positive cancers but development has been discontinued.
