Overview

Peru-15 (Choleragarde) is a live-attenuated, oral cholera vaccine candidate derived from the *Vibrio cholerae* O1 El Tor Inaba strain. Developed by researchers at Harvard University and licensed to Avant Immunotherapeutics (now Celldex Therapeutics), the vaccine was further developed in collaboration with the International Vaccine Institute (IVI). The Peru-15 strain is genetically engineered to be non-toxigenic by deleting the cholera toxin A subunit (ctxA) while retaining the immunogenic B subunit (ctxB). It is also modified to be non-motile and protease-deficient to improve safety and reduce reactogenicity. A trehalose-reformulated version was created to enhance stability for use in resource-limited settings. Clinical trials have evaluated its safety and immunogenicity in adults and infants, particularly in cholera-endemic regions like Bangladesh and India.
