Overview

Pesampator (developmental code names BIIB104 and PF-04958242) is a first-in-class small molecule positive allosteric modulator (PAM) of the AMPA receptor, an ionotropic glutamate receptor. It was originally developed by Pfizer and later by Biogen for the treatment of cognitive impairment associated with schizophrenia. The drug enhances synaptic transmission by potentiating AMPA receptor function, which is thought to improve working memory and other cognitive functions. Pesampator underwent multiple phase 1 trials demonstrating safety and tolerability in healthy volunteers and individuals with schizophrenia on stable antipsychotic regimens. However, development for both schizophrenia-related cognitive impairment and age-related sensorineural hearing loss was discontinued due to insufficient efficacy[1][4][5][7].
