Drug intelligence / Profile preview
Pevifoscorvir (formerly ALG-000184) is an orally bioavailable, small molecule capsid assembly modulator (CAM) of the class-E (empty) phenotype, developed by Aligos Therapeutics for the treatment of chronic hepatitis B virus (HBV) infection. It is a prodrug of the active metabolite ALG-001075. Pevifoscorvir works by binding to the HBV core protein (HBcAg), disrupting capsid assembly, preventing pregenomic RNA (pgRNA) encapsidation, and blocking the establishment and replenishment of covalently closed circular DNA (cccDNA). In clinical trials, pevifoscorvir has demonstrated potent, sustained reductions in HBV DNA, RNA, and viral antigens (HBsAg, HBeAg, and HBcrAg). In 2026, Aligos entered into an exclusive licensing agreement with Xiamen Amoytop Biotech to develop and commercialize pevifoscorvir in Greater China.
