A combination therapy consisting of pexidartinib, a colony-stimulating factor-1 receptor (CSF-1R) inhibitor, and sirolimus, an mTOR inhibitor. This combination targets tumor-associated macrophage (TAM) polarization, particularly depleting M2-type macrophages which are associated with tumor progression. Preclinical studies showed that the combination resulted in increased apoptosis and suppressed signaling pathways (AKT, KIT, and S6 phosphorylation) that neither drug could achieve independently.
