pEZ-IL-27 is a research-grade DNA plasmid used for the expression of Interleukin-27 (IL-27), a cytokine of the IL-12 family, in preclinical studies of epithelial ovarian cancer. In studies using the SKOV3 cell line, transfection with this plasmid leads to the endogenous production of IL-27, which exerts direct anti-tumor effects. The mechanism involves the activation of the STAT3 signaling pathway and the simultaneous inhibition of the Akt pathway, leading to increased caspase-3 expression and induction of apoptosis. Furthermore, pEZ-IL-27 treatment modulates the expression of various cytokines and factors, including the upregulation of IFN-γ, IL-6, and IL-12, and the downregulation of pro-tumorigenic factors such as NF-κB1 and TLR4. It is currently utilized as a tool for investigating gene therapy approaches in gynecologic malignancies.
