Overview

PF-03732010 is a fully human monoclonal antibody developed by Pfizer that targets P-cadherin (CDH3), a membrane glycoprotein involved in tumor cell adhesion, proliferation, and invasiveness. By binding to P-cadherin, the drug inhibits cell adhesion and aggregation mediated by this protein, leading to antitumor and antimetastatic effects in preclinical models. Mechanistically, it reduces β-catenin levels and suppresses mesenchymal-type changes in the cytoskeleton, thereby diminishing metastatic potential. The drug was evaluated in phase 1 clinical trials for solid tumors but was discontinued due to insufficient efficacy[1][2][4][5][6].
