PF-04605412 is a fully human, Fc-engineered IgG1 monoclonal antibody developed by Pfizer that targets the human integrin alpha5beta1 (α5β1). It acts as an antagonist of α5β1 integrin and exhibits dual mechanisms of action with both antiangiogenic and antineoplastic activities. By selectively binding to α5β1 integrin, it prevents the binding of natural ligands such as fibronectin, thereby inhibiting cell adhesion, migration, and endothelial tubulogenesis. Additionally, it elicits potent antibody-dependent cellular cytotoxicity (ADCC) against tumor cells and endothelial cells expressing α5β1. Preclinical studies demonstrated robust single-agent antitumor efficacy associated with macrophage infiltration and increased apoptosis in tumors. The drug was investigated for use in advanced or metastatic solid tumors but development was discontinued after phase 1 trials[1][2][3][4][7].
