Overview

PF-04856883 (also known as CVX-096) is a long-acting glucagon-like peptide-1 receptor (GLP-1R) agonist developed by Pfizer using the proprietary CovX-Body platform. The molecule is a bioconjugate consisting of two GLP-1 mimetic peptides chemically linked to a humanized aldolase monoclonal antibody scaffold. This design utilizes the antibody's long circulating half-life to extend the bioactivity of the GLP-1 peptides, which are otherwise rapidly degraded by dipeptidyl peptidase-4 (DPP-4). PF-04856883 was evaluated in Phase 1 clinical trials for the treatment of type 2 diabetes mellitus, specifically in patients on metformin, but development was ultimately discontinued following Pfizer's decision to close the CovX research site.
