PF-0499132 is a small molecule agonist of the G protein-coupled receptor 119 (GPR119), originally developed by Pfizer for the treatment of type 2 diabetes mellitus. GPR119 is primarily expressed in the pancreatic beta cells and enteroendocrine cells of the gastrointestinal tract. Activation of this receptor by PF-0499132 stimulates the glucose-dependent secretion of insulin and the release of incretin hormones, including glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). This dual mechanism was intended to improve glycemic control and potentially support weight management. However, clinical development was discontinued following early-phase clinical studies.
