Overview

PF-05082566 + rituximab is an investigational combination therapy comprising two agents: PF-05082566 (also known as utomilumab), a monoclonal antibody agonist of 4-1BB (CD137), and rituximab, a monoclonal antibody targeting CD20. PF-05082566 activates 4-1BB, a costimulatory receptor on immune cells, enhancing anti-tumor immune responses. Rituximab selectively targets CD20, a surface protein expressed on B-cells, leading to their depletion through mechanisms such as antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity. The combination is being assessed in patients with relapsed or refractory non-Hodgkin’s B-cell lymphoma, especially follicular lymphoma and diffuse large B-cell lymphoma. Phase 1 trials evaluate this combination’s safety, tolerability, pharmacokinetics, and establish the recommended phase 2 dose[1][3].
